Abstract     Bacteria of the genus Salmonella and species Listeria monocytogenes are important human pathogens that frequently contaminate foods. Classical detection methods for these pathogens require several days (5 - 10) before results are obtained. Inclusion of PCR in detection procedures can facilitate faster testing of food samples. Since there are several problems when using PCR to detect foodborne pathogens (i.e. inhibition of PCR with food components, detection of dead bacterial cells) enrichment of food sample is often used before PCR detection. The purpose of our work was to develop a PCR procedure for simultaneous detection of Salmonella and L. monocytogenes. Specific primer pairs for Salmonella and for L. monocytogenes were used in one amplification reaction and conditions of PCR were optimized. Developed PCR protocol successfully detected both pathogens when concentrations of Salmonella and L. monocytogenes were 105 and 106 CFU/mL, respectively. The protocol could be applied to food samples analyses after enrichment in UPB (Universal Preenrichment Broth).
