Abstrakt     Flow cytometry, used to investigate cells in the flow system instead of on a static microscope, was mainly a research method. From its humble beginnings some 40 years ago, it grew into a technology that is used in a wide range of scientific disciplines. Flow cytometry was available to medicine when reliable diagnosing of immunodeficiency (AIDS) became an essential requirement. Today, the major applications of flow cytometry are hematologic, immunologic and oncologic diagnostics. The emergence of new treatment strategies, based on the use of monoclonal antibodies,is extending the role of cytometry immunophenotyping. Cytometric assays to monitor new generations of targeted therapies are being rapidly developed. STI571 (Gleevec) for chronic myeloid leukemia treatment is one of the first examples of such therapy targeted to known abnormalities in the tumor cells. This report reviews the general principles of flow cytometric immunophenotyping and selected applications of flow cytometry in the clinical laboratory.
