Abstract     The knowledge of the structure and function of certain essential protein inmicroorganism is elemental for the target approach for modelling of new microbial inhibitors. The usual way in this approach is the isolation of the target enzyme and the corresponding gene, purification of the protein and determination of the structure by NMR or X-ray cristalography. The obtained data make possible the use of computer-based techniques which can assist in both the discovery and the optimization of lead compounds. One of the well used bacterial enzyme for drug design is DNA gyrase. This enzyme is involved in several biological processes like initiation of DNA republication, segregation of DNA after replication, gene transription. Inactivation of this enzyme kill the microorganisms. It is known from the literature that N-terminal of the B subunit of the enzyme contains the binding site for certain type of inhibitors A system for expression of the rekombinant 24 kDa N-terminalsub-domain of the DNA gyrase B protein presented in this article.
