Abstrakt | Hydroxysteroid dehydrogenase (HSD) was identified in Shizosaccharomyces pombe and located to the cytosolic fraction as NAD(H) and/or NADP(H) dependent activity. Biotransformation of C 21, and C 19 steroids such as progesterone, 5alpha-pregnane-3,20-dione, 5 beta-pregnane-3,20-dione and their respecftve 3-hydroxysteroids as well as testosterone and 4-androstene 3,l7-dione was examined and all the steroids were found to be substrates. Progestcrone biotraasformation resulted in the reduction of delta 4 double bond of the steroid backbone (in vivo only), as well as the C-3 and C-20 keto groups to give 3alpha-hydroxy-5alpha-pregnane-20-one and 20alpha-hydroxy-4-pregnene-3-one respectively. The C19 steroids, testosterone and 4-androstene-3,17-dione, were interconverted by the same organism. 5alpha-pregnane- 3,20-dione and 5beta-pregnane-3,20-dione were reduced to their 3alpha-hydroxy products. Reverse reactions for oxidoreduction at positions C-3 and C-20, with 3alpha-hydroxy-5beta-pregnane-20-one and 20alpha-hydroxy-4-pregnene-3-one as substrates, were also detected. | |
