Abstract     To gain more information on the muscle energetics we studied the respiration rate of mitochondria with EPRoximetry using the spin probe PAMSL (3-karbamoyl-2,2,5,5-tetrametyl-3-pirroline-1-oksyl). This method is convenient for measuring the respiration or concentration of oxygen in small samples (15 ul). We have measured the influence of DNP and ADP on respiration rate of mitochondria isolated from liver and skeletal muscle of rat. Spin probe molecules dissolved in the sample are sending the local oxygen concentration via the exchange interaction with the paramagnetic oxygen. Therefore the line widths of the spin probe EPR spectra are broadened proportional to the oxvgen concentration. An empirical line shape factor K being proportional to the oxygen concentration has been introduced. Experimentaly it was sllown, that the relation between K and oxygen concentration is not linear and it is also dependent on several conditions of the environment in which the spin probe and oxygen are dissolved. To avoid this experimental problems we reconstruct the spectra theoretically from the spin Hamiltonian and calculate K (CO2).
