Abstrakt | The advent of recombinant DNA technology has generated an interest in peptide and protein drugs. Presently, most of them are administred by parenteral route due to their poor oral bioavailability. The intherent discomforts associated with the parenteral therapy has prompted investigations into other nonparenteral routes and drug delivery techniques. The goal is enhancement of acceptance and compliance by patients, increase of bioavailability and therapeutic index as well as reducing side effects. Rate controlled delivery can be achieved using biopolymers, osmotic minipumps, microencapsulation of secretory cells and other technologies. An opportunity for targeted delivery of proteins is given by antibodies as natural targeting device and colloidal carrier systems such as liposomes, micro and nano particles. Due to comprehension of the problem multidisciplinary effort is needed to solve it. | |
Izvleček | Posledica razvoja rekombinantne DNK tehnologije je veliko število zdravilnih učinkov peptidne narave. Večinoma jih apliciramo parenteralno, saj je biološka uporabnost po peroralni aplikaciji prenizka. Zaradi znanih težav, ki spremljajo parenteralno aplikacijo, se razvijajo novi sistemi. Z njimi želimo nadomestiti invazivno injiciranje, povečati biološko uporabnost in terapevtski indeks ter zmanjšati stranske učinke. Obetavni sistemi z nadzorovano hitrostjo sproščanja zdravilne učinkovine so biopolimeri, osmotske črpalke in mikrokapsulirane sekretorne celice. Za ciljano terapijo in povišanje terapevtskega indeksa kot prenašalce uporabljamo monoklonska protitelesa in koloidne sisteme, kot so liposomi, mikrodelci in nanodelci. ker je problematika prenosa proteinov na mesto delovanja kompleksna, je za njeno rešitev potreben multidisciplinarni pristop. | |
