Abstrakt     This study examined the ultrastructural changes in pregnant rat liver of animals administered with commercial polychlorinated biphenyls (PCBs). Pyralene 3000 (total 120 mg/kg body weight) was given to Wistar rats by intraperitoneal injection on days 7 and 9 of pregnancy. Control animals were injected with vehicle only. On day 12 after the second administration the rats were sacrificed and selected samples (liver, blood and adipose tissue) were collected and processed for further analyses Šhigh resolution gas chromatography (HRGC) and transmission electron microscopy'. HRGC analysis showed a selective enrichment of penta- (Nos. 105, 110, and 118) and hexa-chlorobiphenyl (No. 138) congeners and depletion of tri- and tetra-chloro congeners relative to the original PCB administered in adipose tissue of treated animals. Electron micrographs of the liver from treated animals revealed that the hepatocyte alteration comprised mainly augmentation of smooth endoplasmic reticulum as well as mitochondrial abnormalities, increased peroxisome numbers and reduced glycogen content. It is plausible to suggest that the observed alterations are mainly induced by enriched coplanar mono-ortho PCB congeners Nos.105 and 118.
Izvleček     Opravili smo ultramorfološko analizo jeter brejih podgan, tretiranih z nizkoklorirano tehnično mešanico polikloriranih bifenilov (PCB) - pyralene 3000. Pyralene 3000 (skupno 120 mg/kg telesne teže) smo podganam poskusne skupine aplicirali intraperitonealno 7. in 9. dan brejosti živali kontrolne skupine so prejele samo ustrezno količino nosilca. živali smo žrtvovali dvanajsti dan po drugi aplikaciji, jim odvzeli vzorce jeter, krvi in maščobnega tkiva ter jih pripravili za nadaljnje analize: visoko ločljivo plinsko kromatografijo (HRGC) in transmisijsko elektronsko mikroskopijo. Z analizo HRGC smo ugotovili, da so bile v maščobnem tkivu tretiranih živali višje vrednosti penta- (IUPAC št. 105, 110 in 118) in heksa-klorobifenov (IUPAC št. 138) ter nižje vrednosti tri-(IUPAC št. 28) in tetraklorobifenilov (IUPAC št. 52, 64 in 66) v primerjavi s sestavo v pyralenu 3000. Z ultramorfološko analizo jetrnih tkivnih rezin smo pri tretiranih živalih ugotovili čezmerno razraščanje gladkega endoplazemskega retikuluma ter še spremembe v obliki mitohondrijev, povečano število peroksisomov in izčrpanje glikogenskih zalog. Sklepamo lahko, da so ugotovljene spremembe verjetno posledica delovanja akumuliranih planarnih pentaklorobifenilov (IUPAC št. 105 in 118) v skupnem PCB-ju.
