Abstract     Protein kinase C (PKC) compose a family of serine-threonine protein kinases that play a key role in processes of signal transduction in modulation of cellular growth, metabolism, differentiation and gene expression. The function of PKC in the pathogenesis of cancer, immune and cardio-vascular system diseases and diseases of central nervous system is known as well. Design of agents for modulation of PKC activity and elucidation of its structure and mechanism of action are currently the most intensive fields of research in medicinal chemisty. They provided a variety of PKC inhibitors and some of them are already undergoing clinical trials.
Summary     Protein kinaze C (PKC) so družina serin-treonin kinaz, ki sodelujejo v procesu prenosa signalov pri uravnavanju celične rasti, metabolizma, diferenciacije in pri izražanju genov. PKC so pomembne v patogenezi rakavih obolenj, boleznih imunskega in kardio-vaskularnega sistema ter centralnega živčnega sistema. Načrtovanje učinkovin za modulacijo aktivnosti PKC in hkratni razvoj poznavanja njene strukture ter mehanizma delovanja so v zadnjem času pomembno raziskovano področje farmacevtske kemije, ki je dalo vrsto inhibitorjev PKC, od katerih so nekateri že v fazi kliničnih preizkušanj.
