Abstrakt     In order to examine whether peroxidases (E.C. 1.11.1.7) are involved in the early response of highly susceptible potato (Solauum tuberosum L.) variety Igor to infection with potato virus Y"T" (PVY"T"), their activities were studied during the first 24 hours following inoculation. Soluble, ionically- and covalently-bound peroxidases were analysed in bottom virus- or mock-inoculated leaves and in upper intact leaves as well as in roots. The effect of virus infection was observed already three hours after inoculation, when the activity of soluble and ionically-bound peroxidases decreased in inoculated bottom and upper intact leaves, while the activity of covalently-bound peroxidases increased. The most significant changes between mock and virus inoculated plants were found 24 hours after infection but only in bottom inoculated leaves when the increase of activity of all types of peroxidases was found. Infection had no effect on peroxidase actvities in roots. We can conclud that the observed changes of leaf peroxidases in infected plants contribute to early events in the compatible interaction of susceptible potato variety and PVY"T". Our results indicate that the changes of leaf peroxidases observed three hours after infection contribute to very early systemic response of susceptible potato variety to PVY"T", while the changes observed 21 hours later appears to be connected with the site of infection.
Izvleček     V pričujoči raziskavi smo poskusili ugotoviti ali so peroksidaze (E.C. 1.1 1.1.7) udeležene v zgodnji odgovor dovzetne sorte krompirja (Solanum tuberosum L, cv. Igor) na okužbo s krompirjevim virusm Y"T". Peroksidazno aktivnost smo merili v prvih 24 urah po okužbi. Topne, ionsko in kovalentno vezane preoksidaze smo analizirali v spodnjih z virusom ali s sokom zdravih rastlin (slepa inokulacija) inokuliranih listih in v zgornjih intaktnih listih ter v koreninah. Vpliv virusne okužbe smo zasledili že tri ure po inokulaciji, ko se je aktivnost topnih in ionsko veznih peroksidaz znižala v spodnjih inokuliranih in zgornjih intaktnih listih medtem, ko se je aktivnost kovalentno vezanih peroksidaz zvišala. Najbolj očitne razlike med slepo inokuliranimi in okuženimi rastlinami pa smo opazili 24 ur po okužbi vendar le v inokuliranih listih, ko se je aktivnost vseh treh tipov perokisdaz značilno povečala. Okužba ni vplivala na peroksidaze v koreninah. Opažene spremembe peroksidaz v listih okuženih rastlin lahko pripišemo prvim dogodkom pri interakciji med dovzetno sorto krompirja in krompirjevim virusom Y"T". Rezultati raziskave kažejo, da spremembe peroksidazne aktivnosti, ki smo jih zasledili 3 ure po okužbi, prispevajo k zgodnjemu sistemskemu odgovoru občutljive sorte krompirja na okužbo s krompirjevim virusom, medtem, ko so spremembe opažene 21 ur kasneje, lokalne, povezane z mestom okužbe.
