Abstrakt     The transplantation of autologous human chondrocytes has been used as a surgical procedure since 1994 and from 1995 also at the Clinic for Orthopedy of the Clinical Center in Ljubljana. Chondrocytes are isolated from few cubic millimeters of patient's healthy cartilage biopsy and cultured in vitro to the desired number. The suspension of chondrocytes is further injected into the injured site. Implanted cells fill the space at the damaged site by producing a hyaline-like cartilaginous matrix. In the thesis we have investigated some of the key factors influencing the biopsy transport, monolayer chondrocyte cultivation and the transport of the final cell suspension prepared for the surgical application. The use of three-dimensional tissue preparations could further improve the transplantation results as well as facilitate the surgical procedures. Chondrocytes were thus cultivated on several scaffolds in a three-dimensional environment. We have also introduced some methods suitable for the evaluation of the tissue cultures. We believe that under certain circumstances allogeneic cells could replace the autologous chondrocytes in transplantation procedure. Therefore, we have developed a dendritic cellbased method for the evaluation of the immune response in vitro that could be potentially also applied to the evaluation of the autoimmune reactions.
