Abstrakt | We analyzed the effects of USA standard agricultural practice on the heterogeneity of nitrous oxide reductase gene fragments in native and cultivated soil. We determined the size of representative soil sample (3 g) and isolated total microbial DNA from composite samples of soil aggregates. Cloned fiagments were screened by restriction fragment length polymorphism and sequenced.Various ecological indices were caluclated (Shannon (S), Eveness (E), coverage (C), mean taxonomic distance (D), nonparametric estimator Chaol), Collector curves were plotted for RFLP data and phylogenetic analysis was performed for sequenced nosZ gene fragments. The results showed higher diversity and eveness in cultivated soil than in natvie soil. T-RFLP analysis of marker genes nosZ, 16S rRNA archea and 16S rRNA bacteria has revealed a shift in microbial community from dominant groups in native soil to more even distribution in cultivated soil. The differences in agricultural practice are most probaly responsible for the observed shifts. The native and cultivated soil are inhabited by functionally diverse denitrifying and common microbial communities that cause enhanced nitrous oxide emissions from cultivated soil. | |
