Abstrakt     Original strains of Ganoderma applanatum and Ganoderma lucidum were isolated from Slovenian forests and examined. Triterpenoids from the fruiting bodies of G. applanatum were isolated and determined. G. lucidum was cultivated in a liquid substrate based on potato dextrose and olive oil. The influences of inoculum and oxygen partial pressure in fed batch cultivation in a 10 1 laboratory stirred tank bioreactor were studied. Fungal biomass was found oxygen and shear sensible. Using a 17% (wet weight) 6 days old vegetative inoculum, up to 17.0 gl-1dry fungal biomass was obtained in a fed batch process. Extracellular (1.7 gl-1) and intracellular (0.45 gl-1) polysaccharide fractions were isolated. Polysaccharides were further fractionated and separated by ion-exchange, gel and affinity chromatography. The isolated polysaccharides were mainly (3-Dglucanes. Immunostimulatory effects of one extracellular and four intracellular polysaccharide fractions were tested in vitro on the induction of cytokine synthesis (TNF-alpha, IFN-gamma) in primary cultures of human mononuclear cells (PBMC) isolated from a buffy coat of healthy donors. The results proved the induction of moderate amounts of TNF-alpha (< 3.0 pg mL-1 to 630 pg ml-1 of a culture supernatant), which is comparable to the activity of romurtide, a supporting drug in cancer therapy.
