Abstrakt | Anorexia nervosa and bulimia nervosa are complex diseases, classified as eating disorders. As well as eating, body weight and body image are disturbed. Twin studies are important and show a strong genetic influence in the etiology of the two studied eating disorders. Currently the best model for understanding the etiology of eating disorders is the influence of psychosocial stress on genetically susceptible or weak individuals. The involved genes might influence the way we react to stress, endocrine functions, metabolical states, appetite and personality. The polymorphisms we analysed lie in the genes S-HTT, 5-HTI B, 5-HT2C, COMT, agouti, ghrelin, BDNF, UCP2, DRD2, DRD4 and CHRM3 which belong to serotonin, dopamine and other systems that influence the CNS and metabolic pathways. Results of various analysis between genes and eating disorders are sometimes found to be very contradictory in literature. For example, the polymorphism Met158Va1 in the COMT gene was found to be statistically significant (p = 0,023) in the etiology of AN in Israeli patients (Frisch et al., 2001), but a European study conducted by Gabrovšek et al. (2004) could not confirm this result. Similar results can be found for almost all genes analysed in this thesis. In the Slovene population of patients with AN or BN, we first tested the differences in age of onset and minimum body mass index between the two groups (t-test). Both A00 and BMI were found to se statistically significant between AN and BN patients (A00: p = 0,024; BMI: p = 0,001). We then performed a TDT analysis of markers in family trios and found a statistically significant correlation between COMT and AN (p = 0,0054). We couldn't find any other correlation between AN, BN and any of the other studied alleles. (Abstract truncated at 2000 characters). | |
