Abstrakt | The experimental part of the dissertation was partly done at the Institute of Pharmacology and Experimental Toxicology, Medical Faculty in Ljubljana; the studies of membrane fluidity by Electron Paramagnetic Resonance (EPR) were done at the Jožef Stefan Institute. On the cat model we showed, that antidepressants from different classes after single dosis -we used mirtazapine and citalopram - demonstrate noticeable effects on histamine system. Both drugs prevented the increase of free, applied histamine in the plasma of experimental animal. The neuroleptic propionylpromazine acted similarly. In the research of the research group, in which the candidate was involved, similar effects were demonstrated by tricyclic antidepressant amitriptyline, however, this is the first investigation, where the effects on histamine pharmacokinetics was presumed. When we followed the histamine levels in the whole blood, we found out, that both antidepressants and neuroleptic drug change the distribution of histamine, but we suppose different modes of action. Citalopram predominantly acts on histamine uptake into the blood constituents, in the case of mirtazapinc. the blood cells have less important role. The investigations of membranes of red blood cells have shown, that citalopram causes rearrangement of membrane components, in the way that the membrane fluidity increases, and from increased membrane fluidity we deduced to increased permeability. By following the histamine kinetics in the cat plasma and whole blood and in in vitro experiments we discovered, that the change of histamine pharmacokinetics under influence of mirtazapine, citalopram and mirtazapine can be affected by histamine biotransformation. By the increase of membrane fluidity the effect of intracellular enzymes on histamine catabolism can be augmented. The ascertained »side effects« of the abovementioned psychoactive substances and results about same possible mechanisms are original science contribution. | |
