Abstrakt     BACKGROUND. Clotting factor VIII (FVIII) is a non-enzyroatic plasma protein that acts as an essential cofactor in blood coagulation. Elevated level of FVIII activity is an important and independent risk factor for deep-vein thrombosis and for recurrent thromboembolism. AIM. The aim of our study was to determine the prevalence of elevated FVIII levels in patients with deep-vein thrombosis in Slovenia using the chromogenic method which is more sensitive than the clotting method. HYPOTHESIS. The hypothesis that Slovenian patients with deep-vein thrombosis have elevated activity levels of FVIII, that prevalence of high FVIII activity is higher in patients than in healthy subjects, and that elevated activity level of FVIII is a risk factor for deep-vein thrombosis was tested. SUBJECTS AND METHODS. In the retrospective study we included 45 male and female patients 7 months after deep-vein thrombosis and after their anticoagulation treatment was completed, and 81 sex and age matched healthy controls. Activity of FVIII was determined in plasma samples of all subjects at one occassion, using the chromogenic method. The data have been statistically analysed by two-tailed Student's t-test and by chi-square test. Correlation was calculated by Pearson's correlation test. RESULTS. FV III activity was 168 +- 70% (mean - standard deviation) in the control group and 272 +- 87% in the patient group (p<0,001). The cut-off value of FVIII activity (90th percentile of FVIII activity of the control group) was 254% (95% confidence interval: 239 to 270%). The number of patients with FVIII activity abovc the cut-off value was 26 (58%), which was significantly higher than the number of control subjects with FVIII activity above the cut-off value (N=8, 10% by definition of 90th percentile, p<0,001).
