Abstrakt     Background: More then ten percent of patients who develop breast cancer before the age of 40 are likely to have inherited a deleterious mutation of BRCA genes. To investigate the interaction between the risk factors and genetic susceptibility the international study Case Only Stucdy on the Interaetion of Diet and Genetic Predisposition in the Oeeurrence of Breast Cancer in Young Women was initiated. Slovenian patients represent approximately 10% of internationally pooled data. Objectives: The first objective of this analysis was to predict the number of new breast cancer patients (younger then 40) in Slovenia until 2010. Further purpose was to study the probability of BRCA mutation and the influence of life style factors on the mutation penetrance in Slovenian breast cancer patients diagnosed between 1995 and 2002 before the age of 40 and included into the international research. Methods: The incidence was predicted according to the short term prediction model by Dyba and Hakulinen. To compute the probability of BRCA mutation new software, based on Parmigiani method and named COS-software, was developed. The case-only methodology was applied to study the influence of life style risk factors on BRCA mutation penetrance. The multivariate logistic regression was used to calculate the independent effects of the individual risk factors. Results und conclusions:.Until the year of 2010, the number of breast cancer patients, younger than 40 years, will not significantly change. Currently, the most appropriate software tool for the estimation of the magnitude of genetic susceptibility is COS-software. There was no statistically significant influence of life style risk factors on BRCA mutation penetrance found in our analysis. Due to small pool of slovenian patients and low relative risks in exposition to the life style factors the analysis of of complete european study are needed for more reliable results.
