Abstrakt     Background. Calcium (Ca2+) channel blockers are a group of drugs most often used in cardiovascular diseases. Contributing to their clinical use is also their protective role in hypoxic events. Cerebrocrast and IOS-7531 are novel dihidropyridine derivatives, the most widely used group of Ca2+ channel blockers. AIM. The aim of our work was to study the effects of cerebrocrast and IOS-7531 on contraction of isolated porcine left anterior descending coronary artery (LAD) and compare these effects with those of lacidipine. We also evaluated the effect of cerebrocrast on relaxation of pre-contracted LAD rings after 60-minute hypoxia and 30-minute reoxygenation (HR). Relaxation preserved after HR would imply a protective role on vascular endothelium during hypoxia. Hypothesis. We expected: 1) cerebrocrast and IOS-7531 are both equally effective in inhibiting KCl induced contraction of isolated porcine LAD when compared to lacidipine; 2) cerebrocrast preserves the endothelium-derived hyperpolarization factor (EDHF) mediated relaxation of pre-contracted rings after HR. Methods. Isolated LAD rings (3-4 mm wide) were placed in 10 ml organ baths, filled with Krebs-Henseleit solution at 38 °C. Rings were oxygenated (95% O2 + 5% CO2) in normoxic experiments or nitrogenated (95% N2 + 5% CO2) in hypoxic experiments. In the first series of tests, contractile responses before and after 30-minute incubation with cerebrocrast (1 nM10 microM), IOS-7531 (1 nM-0.1 mM) or lacidipine (0.1 nM-0.1 mM) were measured. After incubation, KCl induced contractions of rings were expressed as percentages of contraction of the same rings before incubation. pD2 (-log IC50) values were determined and compared.
