Abstrakt | The focus of our present study was the regulatory effects of noradrenaline (NA) and serotonin (5-HT) on BDNF synthesis in astrocytes. Examined neurotransmitters potently increased BDNF content in cultured astrocytes in dose- and time-dependent manner: NA (1 miM; 6 h) by 4-fold and 5-HT (1 miM; 4 h) by 2.3-fold in cortical astrocytes and by 2.8- (1 miM; 6 h) and 1.7-fold (0.5 miM; 4 h) in cerebellar astrocytes. Phentolamine (alfa-adrenergic antagonist) showed no effect whereas sotalol (beta-adrenergic antagonist) dose-dependently inhibited the stimulatoty response on BDNF induced by NA, thereby supporting the conclusion that NA-stimulated BDNF synthesis in astrocytes is a beta-adrenergic receptor mediated process. Our results thus indicate the existence of a positive feed-back loop between the activity of monoaminergic neurons and astrocytic neurotrophic support. | |
