Abstrakt     Background. Bladder cancer development is poorly understood, although this is a common desease with it's incidence rising. We therefore analysed properties of histologically well differentiated, noninvasive papillary tumours, that represent early stages in the beginning of cancerous transformation in bladder urothelium, on a cellular as well as on a molecular level, and compared it to control tissue. Methods. SCAN analysis and immunohistochemical evaluation of UPs, urothelial cell differentiation markers, enabled us to assess differentiation state of superficial urothelial cells. Proliferation in urothelium was analysed by immunohistochemistry of PCNA and Ki67 markers. Apoptotic cells were determined by detecion of active caspase 3. The presence of tPA, one of the proteins secreted by the urothelium, was also detected immunohistochemically. On a molecular level, deletions in the three genes on chromosome 9 (CDKN2A, GSN, DBCCR1) and in TP53 gene were tested by real-time PCR. Izvleček     Izhodišča. Razvoj karcinoma sečnega mehurja je slabo pojasnjen, čeprav gre za pogosto bolezen, katere incidenca narašča. V raziskavi smo zato na celičnem in na molekularno genetskem nivoju analizirali lastnosti neinvazivnih papilarnih tumorjev sečnega mehurja, ki predstavfjajo zgodnjo fazo v procesu rakaste preobrazbe urotelija v tem organu, in jih primerjali s kontrolnim tkivom. Metode. S pomočjo vrstične elektronske mikroskopije in imunohistokemijske analize uroplakinov, označevalcev diferenciacije urotelijskih celic, smo določali stopnjo diferenciacije površinskih urotelijskih celic. Z imunohistokemijsko analizo označevalcev PCNA in Ki-67 smo ugotavfjali proliferacijsko aktivnost urotelija. Apoptotske celice smo določali z metodo zaznave aktivne kaspaze 3. Prav tako smo imunohistokemijsko zaznavali prisotnost tkivnega aktivatorja plazminogena, enega izmed proteinov, ki jih urotelij izloča. Na molekularno genetskem nivoju smo s pomočjo verižne reakcije s polimerazo v realnem času testirali prisotnost delecij v treh genih na kromosomu 9 (CDKN2A, GSN, DBCCR1) ter v genu TP53. Verižno reakcijo s polimerazo smo uporabili za analizo delecij sedmih lokusov na kromosomu Y. (Izvleček skrajšan na 2000 znakov)
