Abstrakt | This article describes molecular genetic methods for identifying unidentified human skeletons or strongly decayed human remains. DNA was isolated from the long bones (femur) by a method of organic extraction, without previous decalcification. In the teeth, the pulp was used as a DNA source and extraction was based on the Chelex method. The samples were quantified by a method of real-time quantitative PCR by using a Quantifiler Human DNA Quantification kit. DNA concentrations were within 0.05 ng/micro l and 20 ng/micro l, which was sufficient in all cases for successful multiplication of microsatellite loci of the nuclear DNA with an AmpF/STR IdentifilerTM PCR Amplification Kit and analysis with an automatic ABI PRISM TM 310 Genetic Analyser. We acquired complete genetic profiles, whose peaks exceeded 150 RFU (relative fluorescent units). The results of research in practice have shown that, when identifying strongly decayed bodies and human skeletons, the selection of molecular genetic techniques is very reasonable and also the most reliable identification method. | |
