Abstrakt     Monoclonal antibodies (mAbs) directed against different epitopes on molecules of the prion protein (PrP) are at present, one of the most promissing diagnostic and therapeutic tools for transmissible spongiform encephalopaties (TSE). A panel of mAbs directed against recombinant human PrP was prepared. Isolated spleen lymphocytes from immunised Prnp knockout mice (Prnp0/0 mice) were fused with murine myeloma cells. Hybridoma cell lines producing specific antibodies were screened with indirect ELISA test against recHuPrP. Hybridoma cell lines were then cloned by limited dilution and four specific, very potent mAbu against recHuPrP were obtained. All mAbs belonged to the IgG2a subclass. The cloned antibodies were purified and characterized by Western blot analyses. The differences in immunoblots indicated, that mAbs were directed to different epitopes on human PrP.
