Abstrakt     Background: People and animals with inflamed periodontal tissues have increased tendency to develop pneumonia on aspiration of saliva. Porphyromonas gingivalis (Pg), a major periodontal pathogen, has been isolated from inflamed lungs. Material and methods: Twelve-week-old BALB/c mice were infected with Pg ATCC 33277, administered by intratracheal injection at two different concentrations. The animals were sacrificed at 2, 6, 24 and 168 hours post infection. TNF-alpha, IL-1beta and IL6 were determined in serum, lungs and kidneys. Total protein level was measured in kidneys. Specimens of lung and kidney tissue were processed for light microscopy. Kidney tissue was examined by immunohistochemical staining to detect smooth muscle actin a. Results: In animals infected with the higher concentration of Pg, TNF-alpha levels in serum and kidneys were increased at 6 hours and in lungs already at 2 hours post infection; IL-1beta levels in serum were increased at 2 to 72 hours, in lungs at 6 and 24 hours, and in kidneys at 24 hours; IL 6 levels in serum were elevated at 72 and 168 hours, in lungs at 6 hours, and in kidneys at 2 to 24 hours; total protein levels in kidneys were increased at 6 to 168 hours. In the same group of animals, histological studies of lungs demonstrated peribronchial inflammatory infiltration at 24 hours and severe perialveolar inflammatory infiltration without pulmonary parenchymal necrosis at 72 hours; in kidneys, increased quantity of smooth muscle actin a was observed about glomerular capsules at 24 hours. Conclusion: The results of our study demonstrate that Pg infection in mice elicited local and systemic inflammatory and immune responses. It resulted in the development of inflammatory infiltrates in the lungs and increased deposition of smooth muscle actin a in the kidneys. The latter may be attributed to stimulation of mesangial cells and podocytes by immune complexes present in systemic circulation.
Izvleček     Izhodišče: Ljudje in živali z vnetimi obzobnimi tkivi ob aspiriranju sline pogosteje zbolijo za pljučnico. Raziskovalci so iz vnetih pljuč izolirali različne bakterije iz ustne votline, tudi Porphyromonas gingivalis (Pg), ki je udeležena pri parodontalni bolezni. Material in metode: Uporabili smo 12 tednov stare miši BALB/c, ki smo jim pljuča prek sapnika okužili z dvema različnima koncentracijama (105 ali 109 CFU) Pg ATCC 33277. Okužene miši so bile žrtvovane 2, 6, 24, 72 in 168 ur po začetku poskusa. V serumu, pfjučih in ledvicah smo določali raven TNF-alfa IL-1beta, IL-6 ter koncentracijo celokupnih beljakovin. Preparati pljuč in ledvic so bili pripravljeni za svetlobnomikroskopsko analizo. Na ledvicah smo opravili tudi imunohistokemično analizo na gladkomišični aktin a. Rezultati: Pri višji koncentraciji Pg (109 CFU) so bile vrednosti TNF-alfa v serumu in ledvicah značilno višje po 6 urah, v pljučih pa že po 2 urah. Vrednosti za IL-1beta v serumu so bile značilno višje 2 do 72 ur po okužbi, v pljučih po 6 in 24 urah in v ledvicah po 24 urah. Prav tako so bile vrednosti za IL 6 pri višji koncentraciji v serumu značilno višje po 72 in 168 urah, v pljučih 6 ur po koncu poskusa in v ledvicah po 2 urah ter so bile višje do 24 ur. Ob višji koncentraciji Pg je bila vrednost celokupnih beljakovin v ledvicah značilno višja že po 6 urah in je trajala do konca poskusa. Histološko smo 24 ur po okužbi v pljučih ugotovili vnetno infiltracijo okrog bronhiolov in po 72 urah močno vnetno infiltracijo okoli alveolov, brez parenhimske nekroze pljuč. V ledvicah smo okrog kapsule glomerulov po 24 urah ugotovili povečano količino gladkomišičnega aktina a. Zaključek: Dokazali smo, da je okužba s Pg vzpodbudila lokalni in sistemski vnetni in imunski odziv. (Izvleček skrajšan na 2000 znakov)
