Abstrakt | Background. Amelogenesis imperfecta (AI) is common name for phenotipicaly and genotypic diverse inherited disorders of dental enamel. There are many different classifications of AI phenotypes; however none of classification entirely meets the requirements. Due to clinical diversity of AI it can be sometimes difficult to classify a certain AI type. There are no reports on correlation between clinical appearance and structure and composition as well as genotype in the literature. Dental enamel microhardness on teeth with AI has not been evaluated until now. Also, the ultrastructure of enamel with ENAM gene mutation g.13185-13186insAG has not been described previously. Methods. Patients with different types of AI: pitted hypoplastic, smooth hypoplastic, rough hypoplastic, hypomaturated and hypocalcified, were included in this research. On axial cuts of AI teeth acquired from the patients enamel ultrastructure was analyzed using scanning electron microscopy (SEM), enamel microhardness was evaluated using the Vickers scale and concentration of elements Ca, P, Mg and Na were measured using energy dispersive spectrometer (EDS). In genomic DNA of the patients we searched for a mutation in enamelin gena (ENAM), and in families with only male affected also for a mutation in amelogenine gene (AMGY) employing sequence analysis of polymerise chain reaction (PCR) products.
