Abstrakt     Histamine (Hi) is released into blood during physiological and many pathophysiological processes. Since its elimination from the general circulation is fast, measurement of Hi in plasma as a marker for Hi release can be unreliable. The aim of this thesis was to study some pharmacokinetic characteristics of Hi and its metabolite, tele-methylhistamine (MeHi), particularly their elimination rate from the circulation. On the basis of these results we also wanted to search a significance or advantages of MeHi measurement simultaneously with Hi or instead of Hi in blood or plasma. The kinetics of Hi and MeHi, and effects of increased Hi concentrations on the levels of MeHi in the systemic circulation were studied in the rat and in the cat. We modified a method for rat plasma preparation and a method for simultaneous Hi and MeHi assay by HPLC from blood and plasma of an experimental animal. The results got us new data about the ratio of both amines in the blood and plasma of the rat and the cat. In physiologic conditions the levels of plasma Hi are only l0 per cent (rat) or 1.2-3.5 per cent (cat) of whole blood Hi level, while concentrations of plasma MeHi represent 25 per cent lrat) or 8.1-14. 3 per cent (cat) of MeHi concentrations in the whole blood. In cat plasma MeHi levels are higher than Hi levels. An increase of Hi levels (either exogenous or endogenous) in the circulation of experimental animals was followed by an increase of MeHi in blood and plasma. In the rat an intravenous injection of histamine liberator (comendogenous Hi and MeHi levels in plasma. nation of Hi. pound 48/80) caused an enormous increase of endogenous levels of blood Hi reaching maximum within 10-15 min. The levels of newly formed MeHi reached maximum within 40-45 min. (Abstract truncated at 2000 characters).
