Abstrakt     The author has elaborated an accurate and reliable methodological approach for identification and differentiation of autoantibodies in rnonospecific and polyspecific sera of patients with systemic connective tissue diseases against the following intracellular antigens: Sm, U1RNP, Ro, La, SL, Scl-70, Jo-1, PM/Scl, ribRNP and PDH. For this purpose specially designed variation of counterimmunoelectrophoresis (CIE) was developed, which was accepted on the European level as one of the four standard techniques for the detection of autoantibodies. By the proposed methodology for defining autoantibodies in polyspecific sera, it was proven that autoantibodies in sera with different mixed specificities could be precisely determined. CIE is ten times more sensitive than radial immunodiffusion technique. The identification of specific autoantibodies is based on a demonstration of lines of immunological identity with standard antisera and for this reason CIE is often more reliable than more sensitive enzyme-linked immuno-sorbent assays and immunoblot techniques for which recombinant antigens are not always available. With adequate selection of antigen substrates and appropriate application of this methodological approach, a precise detection of above mentioned autoantibodies can be performed, except for autoantibodies against ribosomal ribonucleoproteins, which can be more accurately identified by anti-P enzyme-linked immuno-sorbent assay and immunoblot on either ribosomal preparation from HeLa cells or ribRNP/anti-ribRNP immune complexes precipitated in CIE gel. The most suitable antig en substrate for the detection and differentiation of autoantibodies against Sm, UlRNP, La, SL, Scl-70, Jo-1 and PM/Scl by CIE is an extract from rabbit thymus acetone powder.(trunc.)
