Abstrakt     The proliferative activity of different thyroid tumours was evaluated with three proliferation markers: Ag-NOR, PCNA and MIB-1. The aim of the study was to determine if any of these markers, used alone or in combinations of two, could be of value in differential diagnosis of thyrocyte tumours. The efficiency, specificity, sensitivity and predictive value of negative and positive results were determined for each proliferation marker used. Fifty-four thyrocyte tumours were analysed. They were divided into six groups: simple, hyperactive and oxyphilic adenomas and follicular, papillary and anaplastic carcinomas. Each group included 10 tumours except the group of anaplastic carcinomas which comprised only 4. Histological sections were impregnated with silver for Ag-NOR visualisation or stained immunohistochemically with anti-PCNA or MIB-1. With the CAS 200 and IBAS 1000 image analysing systems mean thyrocyte nuclear areas, perimeters and maximal diameters, the standard deviations of the nuclear area, the numbers of Ag-NOR per nucleus, the percentages of nuclei with 5 or more Ag-NORs, and the percentages of PCNA and MIB-1 positive nuclear area were determined. The results for different groups were compared by multivariate analysis. Correlation between individual variables and the metric characteristics for each variable were defined. The best combinations were defined. The best combinations were estimated with logistic regression analysis. The results showed significant differences between simple adenomas and thyrocyte carcinomas with respect to the number of Ag-NORs per nucleus and the percentage of nuclei with 5 or more Ag-NORs. There were also significant differences between anaplastic carcinomas and all others groups of thyrocyte tumours regarding the number of Ag-NORs per nucleus, the percentage of nuclei with 5 or more Ag-NORs, and the percentages of PCNA and MIB-1 positive nuclear area.(trunc.)
