Abstrakt     The electrophysiological patch-clamp technique was applied to study the secretory activity of a single cell by measuring changes in membrane capacitance. This parameter is related to changes in surface area which reflects membrane cycling processes such as endo-and exocytosis. In this paper a brief outline is given on the progress of understanding the mechanisms controlling exocytosis in a single adenohypophyseal cell. Since the cytosol composition of a single cell can be controlled by the patch-clamp technique, by dialysis from the patchpipette, the influence of various bioactive molecules and second messengers on the secretory apparatus can be studied. Moreover, by cytosol dialysis with antisense DNA oligo probes, thus blocking the translation of a specific protein, the role of GTP-binding proteins in the stimulus secretion coupling can be studied.
