Abstrakt     Ten years of experience with an ELISA test using deoxyribonucleoprotein (DNP) as an antigen is presented. The correlation of the results with the classical LE-cell test is highly significant, accordingly a common molecular basis for both tests may be assumed. Specificity for systemic lupus erythematosus (SLE) is between 80-90 percent and sensitivity exceeds 92 percent in untreated SLE patients. In the longitudinal studies, the anti-DNP levels vary in individual patients, falling to low levels during low activity of the disease or in remission. Preparation of the DNP antigen is rather simple and the variability of the test results in long-term studies can be kept within acceptable limits to provide objective, reliable and quantitative results. Anti-DNP-ELISA may be an alternative to the LE-cell test, enabling the estimation of the first recognized autoantibody in SLE: the LE-factor (Hargraves,1948).
