Abstrakt     Background. The reticulocyte count is a clinically important indirect indicator of erythropoietic activity in patients. However, determination of the reticulocyte count using a light microscope has low accurracy and reproducibility rates. Recently, automated reticulocyte counters have been developed, which allow for a more rapid and accurate counting of reticulocytes. Methods. In our study, we compared the following methods for reticulocyte count determination: the microscopic brilliant cresyl blue method and flow cytometric EPICS Profile (Coulter) method using thiazole orange (TO). The purpose of the study was following: to select the most suitable TO concentration to be used with the EPICS Profile cytometer. The following TO concentrations were used: 0,2 mg/L, 0.1 mg/L, 0.07 mg/L 0.05 mg/L. Results and conclusions. The most appropriate TO concentration for counting reticulocyte by the EPICS Profile is 0,1 mg/L. The concentration was select according to the following criteria: good agreement of the results obtained by microscopy with the results yielded by the EPICS Profile cytometer, absence of statistically significant differences between the results, and agreement in the normal ranges. There was a good correlation between the methods studied. The correlation coefficent (r) ranged from 0,82 and 0,87. The mean intra-assay coefficients of variation for determination by light microscopy and EPICS Profile cytometer was 27.5% and 8.4% respectively.
Izvleček     Izhodišča. Število retikulocitov v krvi je klinično pomembno kot posredni kazalnik obsega nastajanja eritrocitov v kostnem mozgu. Določanje števila retikulocitov s svetlobnim mikroskopom je slabo ponovljivo in nenatančno. V zadnjem času so razvili avtomatične retikulocitne števce, ki omogočajo hitrejše in natančnejše štetje retikulocitov. Metode. V nalogi smo primerjali določanje števila retikulocitov s svetlobnim mikroskopom in pretočnim citometrom EPICS profile (Coulter). Za štetje retikulocitov s svetlobnim mikroskopom smo vzorce krvi obarvali z briljantnim krezilnim modrilom, za določanje s pretočnim citometrom pa s tiazol oranžnim barvilom (TO). Želeli smo ugotoviti, katera koncentracija TO je najprimernejša z apretočni citometer EPICS profile. Izbirali smo med naslednjimi koncentracijami TO: 0,2 mg/L; 0,1 mg/L; 0,07 mg/L in 0,05 mg/L. Rezultati in zaključki. Najprimernejša koncentracija tiazol oranžnega barvila za štetje retikulocitov s pretočnim citometrom EPICS Profile je 0,1 mg/L. Izbrali smo jo na osnovi najboljše skladnosti rezultatov med svetlobnim mikroskopom in pretočnim citometrom EPICS Profile, najmanjših razlik med rezultati, ki smo jih analizirali s parnim t-testom in ujemanja normalnih vrednosti zdravih odraslih krvodajalcev. Stopnja povezanosti med dvema načinoma določanja retikulocitov je bila dobra. Korelacijski koeficient (R) je bil med 0,82 in 0,87. Povprečna koeficientna variacije štetja retikulocitov s svetlobnim mikroskopom in pretočnim citometrom EPICS profile sta bila 27,5% in 8,4%.
