Abstrakt     Pharmacokinetics deals with transports of substances in organisms. pharmacokinetic modellings is mostly performed by the aid of compartment models. Many different kinds of drug administration are frequently approximated by a step or pulse function which represents a discontinuity in the input signal. The latter may cause numerical problems for the digital simulation tool if it is not treated correspondingly. The similar can be said for the consequence of model stiffness which is often the case in pharmacokinetic studies. The work deals with the mentioned problems of pharmacokinetic modelling for the case of three antibiotics. Measured data in the case was obtained in the extensive "in vivo" study. The aim was to support the hypotesis that the antibiotics kinetics changes when the patients status changes from fever to defervescence. It came out that the discontuity as well as stiffness problems can be, in numerical sense, very unpredictable. The first problem can be avoided with inclusion of special discrete block in Simulink, while the second on erequests appropriate numerical integration algorithm.
