Abstrakt     Pseudomonas stutzeri JM 300 can express two nitrate reductases. Membrane bound nitrate reductase is induced by nitrate, oxygen tolerant periplasmic nitrate reductase (NAP) is synthesized when the cells grow in a complex medium without nitrate. Specific activity of NAP in the cells increase 10 fold at the end of the exponential growth phase and it is influenced by growth constant of the chemostat culture. If the cells grow in a defined medium specific activity of NAP is influenced by the nature of carbon substrate and it serves as a mechanism by which reductant can be dissipated to maintain the cellular redox state. Some biochemical characterization were made on partially purified enzyme. Apparent molecular weight defined by gel filtration is 85 kDa, pH optimum 6.3 and temperature optimum 60 degrees C. Km for nitrate with methylviologen as electron donor is 0.27 mM. SDS PAGE electrophoresis revealed monomeric structure of the enzyme.
