Abstrakt     A crucial property in the pathogenesis of Shigellae is the microorganism's capacity to invade the cytoplasm of eukaryotic cells and elicit an immune response. The bacterial entry into the cytoplasm of macrophages can be correlated with the induction of apoptosis. On the other hand, invasive wild-type strain of S. flexneri (M90T) and a plasmid cured noninvasive derivative (BS176) remain entrapped in the phagolysosome of PMN, as visualized by electron microscopy. Both strains are killed by this phagocyte by an oxigen independent mechanism with similar efficiency. One of the PMN proteins responsible for the killing is bactericidal/permeability increasing protein or BPI (Mandić-Mulec, et al., 1997). To understand why Shigellae do not enter the cytoplasm of PMN we examined the effects of protein extracts from PMN on invasive properties of S. flexneri. Our results suggest that S. flexneri pretreated with PMN extracts is able to induce apoptosis in macrophages but the entry into programmed cell death of infected macrophages is delayed as compared to macrophages infected with untreated Shigellae.
