Abstrakt     Paratuberculosis is a chronic infectious disease of ruminants caused by Mycobacterium avium subsp. paratuberculosis. The disease is important because of great economical losses caused in infected herds. The diagnosis of paratuberculosis is difficult because of the lack of accurate and sensitive diagnostic test to detect all infected animals into the herd. We have compared four serological methods for the diagnosis of paratuberculosis in sheep: complement fixation test (CF), agar-gel immunodiffusion test (AGID), enzyme-linked immunosorbent assay (ELISA) with preabsorption with suspension of Mycobacterium phlei and ELISA without preabsorption. Altogether we have tested 1365 sheep sera originated from the flock with paratuberculosis. In addition we have olso pathomorphologically and bacteriologically examined 70 sheep that died in the time of survey. In ELISA without preabsorption 6,6% sera were positive and 11,2% inclonclusive, in ELISA with preabsorption 2,1% sera were positiven and 2,6% inconclusive. In CF test 0,2% sera were positive and 1,9% inconclusive and in AGID test 0,4% sera were positive and 0,1% inconclusive. Pathomorphologically 11 animals were positive and 7 animals were inconclusive. Causative agent was isolated from 3 animals. When comparing the results of all four test we can conclude that ELISA with preabsorption scems to be the best method for the diagnosis of paratuberculosis in sheep.
