Abstrakt     Diagnostics of leptospirosis on post-mortem tissues is not always successful, thus inducing the need for the developing of new methods. Beside standard histological methods for detection of leptospiral microorganisms in tissues, like silver staining, it is possible to use immunohistochemical techniques.For this approach we need specific antibodies that selectively react with leptospiral antigen. The aim of our work was to produce polyclonal antibodies against serovars of Leptospira interrogans most often found in Slovenia. We immunised rabbits with nonpathogenic serovars of Leptospira interrogans serovar pomona, serovar icterohaemorrhagiae and serovar grippotyphosa. Antibodies were tested on tissues of animals serologically and bacteriologically positive for leptospirosis. Tissues were fixed in buffered neutral formalin, and paraffin embedded. The procedure with labeled streptavidin biotin (LSAB, Dako) was used for the immunohistochemical demonstration of antigen, and reference antibodies and tissues positive for immunohistochemical demonstration of antigen, and reference antibodies and tissues positive for different types of leptospires as a control. Immunohistochemical reaction at the location of leptospiral antigen in the tissue, obtained with the antibodies that we have produced, was strong and cpecific for leptospiral antigen, but not entirely specific for individual serovars.
