Abstrakt     Background: This article focuses on prevalence of animal dermatophytoses in Slovenia and corresponding diagnostic techniques (Wood's lamp examination, light, and fluorescent microscopic examination of scales, hairs, and fungal culture). A comparison of protein profiles among different species and among Microsporum canis strains was performed as well. Materials and methods: In the period from January 1996 to February 1997 mycological examinations of 436 skin scrapings and hair samples of pet animals were made. The majority of samples were obtained from cats (75%) the rest from dogs (25%). All the samples were analyzed by cultural examination on two different media: Sabouraud dextrose agar with penicillin and streptomycin (SDA) and Sabouraud dextrose agar with chloramphenicol and actidion (SCA). Direct microscopic examination and fluorescent microscopic examination were performed as well. Protein profiles of cytoplasmic extracts from disrupted mycelium of dermatophytes were compared by polyacrylamide gel electroporesis in presence of sodium sulphate (SDS PAGE). Results and conclusions: Dermatophytes Microsporum canis, Trichophyton mentagrophytes and Microsporum gypseum were isolated. Although a number of examinations are available, the fungal culture is still the most reliable diagnostic method. Sabouraud agar with chloramphenicol and actidion is the most suitable medium. Wood's lamp examination and both microscopic techniques were primarily used for further confirmation of suspicious hair shafts. Protein analysis, using gradient gel with 8-25% of acrylamide showed small and inconsistent differences in protein pattern among M. canis. A comparison of protein profiles among different dermatophyte species (M. canis, M. gypseum, T. mentagrophytes, T. verrucosum) showed more evident differences. Therefore the SDS-PAGE method for analysis of dermatophyte protein patterns can be used for the identification of species.
