Abstrakt     A method is described for the determination of residues of dimetridazole in muscle tissue. After the tissue has been homogenized in dichloromethane, the extract is evaporated and redissolved in hexane. Dimetridazole is extracted into a buffer solution. Further purification is done with SPE on C-18 column. Dimetridazole is eluted with a buffer and methanol mixture, concentrated and redissolved in the mobile phase. Quantification is done with HPLC on reversed phase column (C-18) using acetonitrile, water, acetic acid and triethanolamine as mobile phase with UV detection at 320 nm. The limit of detection is 5 microg/kg and the limit of quantification is 10 microg/kg with the coefficient of variation 11.8 % . Linear calibration graph in matrix was obtained in the range 5-50 microg/kg with the correlation coefficient 0,9997. The situation regarding residues of dimetridazole in Slovenia from 1991 to 1997 is shown.
Izvleček     Opisali smo metodo ugotavljanja ostankov dimetridazola v mišičnih tkivih. Dimetridazol izločimo iz vzorca z diklormetanom, čemur sledi uparitev ekstrakta. Dobljene ostanke raztopimo v heksanu, izločimo v pufrsko raztopino in nadaljujemo čiščenje na kolonah C-18. Dimetridazol eluiramo iz kolon z mešanico pufra in metanola. Dobljeni eluat koncentriramo, ostanke pa raztopimo v mobilni fazi. Končno vrednotenje opravimo s tekočinsko kromatografijo na koloni C-18 ob uporabi mešanice acetonitrila in acetatnega pufra pH 5 kot mobilne faze in z meritvami absorbance pri 320 nm. Meja zaznavnosti je 5 microg/kg, meja vrednotenja 10 microg/kg z relativnim standardnim odklonom 11,8 %. Umeritvena krivulja v vzorcu je v območju od 5 do 50 microg/kg linearna s korelacijskim koeficientom 0,9997. Prikazali smo stanje v zvezi z ostanki dimetridazola v Sloveniji od leta 1991 do 1997.
