Abstrakt     Determination of free fatty acids (FFA) and beta-hydroxybutyrate (BHB) was performed on automatic biochemical analyser Cobas Mira and repeatability, reproducibility and accuracy of measurements were determined. The repeatability was good at normal and high level of FFA in blood serum (CV % 3.25 and 0.86 %), and a bit worse, but still acceptable at low levels (CV % 4.55). The repeatability for BHB determination was good at low (CV % 2.47), normal (CV % 1.36) and high levels (CV % 0.65%). Reproducibility was examined with two control serum samples at normal and high levels and was satisfactory for both analyses. The accuracy of measurements was determined by adding different concentrations of standard solutions to blood serum samples. Before estimating the regression between measured and added concentrations, the endogenous level of FFA and BHB were subtracted. The results were good. In concluiosn the introduction of new analytical methods expanded the possibility of early detection of subclinical and clinical metabolic diseases as well as the extent of the metabolic profile in ruminants.
Izvleček     Vsebnost prostih maščobnih kislin (FFA) in beta-hidroksibutirata (BHB) smo določali na avtomatskem biokemijskem analizatorju COBAS MIRA in ugotavljali prciznost, ponovljivost med dnevi in natančnost meritev. Ugotovili smo, da je bila preciznost dobra pri srednji in visoki vsebnosti FFA (CV % 3,25 in 0,86), nekoliko slabša, vendar še sprejemljiva, pa pri nizki vsebnosti (CV % 4,55). Preciznost meritev pri določanju BHB v krvnem serumu je bila dobra pri nizki (CV % 2,47), srednji (CV % 1.36) in visoki vsebnosti (CV % 0,65). Ponovljivost meritev med dnevi smo preverjali z dvema kontrolnima serumoma na normalni in visoki ravni in je bila zadovoljiva pri obeh analizah. Natančnost meritev smo preverili z dodajanjem različnih koncentracij standardnih raztopin krvnemu serumu, ki smo mu predhodno izmerili endogeno vsebnost FFA in BHB. Rezultati so bili dobri. Z novima analitičnima metodama smo razširili možnost zgodnjega odkrivanja subkliničnih in kliničnih presnovnih bolezni in obsega metaboličnega profila pri prežvekovalcih.
