Abstrakt     Potato tuber necrotic ringspot disease, caused by necrotic strain of potato virus PVYNTN, is one of the most harmful diseases that infected Slovenian potato crops in the last century. In order to obtain successful system for the potato Solanum tuberosum cv. Igor and to develop virus-resistant transgenic plants, the transformation plant technique with insertion of gene was used. The capability of regeneration and calli formation on potato plant Solanum tuberosum cv. Igor was observed on the media with different concentration of the kanamycin. The results of the study concluded that shoot growth is the least obstructive with 50% decreased concentration of kanamycin. The tobacco plants Nicotiana tabacum cv. Samsung and the potato plants Solanum tuberosum cv. Igor were infected with Agrobacterium tumefaciens. Therefore, a gene block with coat protein (CP) gene of virus PVYNTN - in sense orientation or gene which coded unfunctional protein and also a gene for kanamycin selection were introduced in Agrobacterium tumefaciens strain. During the process of Nicotiana tabacum transformation, an increased virus-resistance and stable transgenic lines were developed, which has been preserved in tissue culture with micropropagation technique and was afterward transferred to the soil in a greenhouse. The highest ratio of calli formation, the highest number of shoots and the highest transformation was achieved on internodie explants. Furthermore, the transformation of potato Solanum tuberosum cv. Igor obtained more stabile transformed plants as well, which were then used for further tests of resistance against PVYNTN virus.
