Abstrakt     Introduction: Geographic variations in the survival of patients with thyroid carcinoma are well known. The reasons could be the differences in the distribution of histologic types, possibly also DNA ploidy and proliferative activity (PA). The aim of this study was to determine the ploidy and PA in thyroid carcinomas in Slovenia in order to provide a base for future studies of prognostic factors. material and methods: DNA ploidy and PA were analysed in 202 patients (43 men, 159 women, mean age 55,5, range 14-91) with primary thyroid carcinomas (89 papillary, 19 medullary, 45 follicular and 49 anaplastic) treated at the Institute of Oncology from 1988 to 1999. Flow cytometric DNA measurements were performed on Partec PAS I ans PAS II mercury lamp flow cytometer on fresh fine needle biopsies from primary tumors. DNA was stained with fluorochrome DAPI, proteins with SR101(mean CV was 2,5, range 0,8-6,65, mean number of signals was 18900). All samples were analysed with internal standard composed of normal stromal cells amd leukocytes. In 176 tumors, in addition to internal standard, an external standard with human lymphocytes and/or throut erythrocytes was used. The results of the two methods of standardisation were compared. Proliferative activity was determined in all diploid and in aneuploid carcinomas with separated cell peaks. Results: Aneuplody (tetraplody included) was found in 24% (21/89) of papillary, 47% (9/19) medullary, 67% (30/45) follicular and 73% (36/49) anaplastic carcinomas.
