Abstrakt     This study investigates the kinetics of immunologic reactions between triiodothyronine (T3), thyroxine (T4), thyroglobuline (Tg) and their animal and human antibodies, especially the antigen/antibody association and the dissociation of immune complexes. Specific antisera against T3 and T4 were prepared by the immunisation of rabbits with T3-BSA and T-BSA conjugates, respectively. Low titres of antisera against T4 (antiT4) found, were dur to binding of endogenous rabbit T4 to antiT4. Rabbit antiT4 titres were increased by coupling antiT4 to the T4-epoxy-activated Sepharose 4B conjugate and by the elution of these immunoadsorbents with guanidine chloride 3 Mol/l or ammonium hydroxide 1 Mol/l. The use of ammonium hydroxide for the dissociation of immune complexes is new one, its advantage being simple elimination of ammonium ions from water solutions, leaving the antibodies intact. The T4/antiT4 reaction depends on pH of the medium used. Optimal binding occurs at pH 3-7, but the addition of normal human serum, shifts it to pH 3-4. pH of the medium, however, seems to have no notable influence on antisera against T3. On testing thyroid patients for the presence of endogenous antiT3 and antiT4, antiT3 were found in two cases, and antiT4 in none. Apart their influences on the results of the T3 and T4 immunoassays. The role of endogenous antiT3 and antiT4 in the course of autoimmune thyroid diseases is still poorly understood. The study of autoimmune antobodies against Tg (antiTg) involved inestigations of possible irreversible Tg-antiTg immune complexes which can not be measured using standard immunoassay techniques. Various methods for the disociation of Tg-antiTg complexes were studied in this connection. It was found that 8-anilino-naphtalene-sulfonic acid (ANS) inhibits the Tg/antiTg reaction. On the basis of this new finding a procedure for previous treatment of serum samples was developed using ANS and dialysis.(trunc.)
