Abstrakt		In prospective comparative open study the kinetics of plasma levels of pefloxacin and urine excretion of pefloxacin with its metabollite norfloxacin was investigated. The once-daily dosage regimen was designed: 400mg oral pefloxacin (Abaktal) was administered daily for 7 days. Ten healthy volunteers participated in the study. Pefloxacin was administered at 7.30 a. m. in regimen 1 and 6 p. m. in regimen 2, pefloxacin plasma concentrations were 4.25mg/l-6.02mg/l (1.5 hour after the dose) and 0.86mg/l-1.61mg/l (just before the next dose). After 24 hours 102.56mg-109.34mg of pefloxacin and 95.82mg-101.48mg of norfloxacin was excreted in urine. The dose of 400mg of pefloxacin in once-daily dosage regimen is suitable for major lower as well as upper urinary tract infections treatment if bacteriemia is excluded. Such treatment regimen is useful when period of severe illness is overcome
