Abstrakt     The pharmacokinetics of piroxicam following oral administration was examined in male rats with a CCl4-induced liver injury and compared with control rats. The fasted animals were pretreated orally with CCl4 in 0.05 and 1.0 ml kg-1 doses and after 24 hours piroxicam (20 mg kg-1) was administered. The control animals were given piroxicam only. Blood samples collected up to 96 hours and piroxicam plasma levels were determined by the spectrofluorometric method. The assessment of liver injury was made histologically 24 hours after CCl4 administration. in both CCl4-treated groups significantly higher (p < 0.05) plasma piroxicam concentrations were found, especially in the higher CCl4 dose group. The calculated pharmacokinetic parameters demonstrated great differences between the control rats and those rats with liver injury especially expressed in the higher CCl4 dose group. The values of clearance and volume of distribution show considerable reduction and were: 7.4, 2.24, 1.08ml h-1 and 102.57, 65.16 and 61.36 ml for the control, low and higher CCl4 dose groups respectively. Biological half-lives were prolonged to 16.44h (low dose) and 39.55h (high dose) in comparison with the control group (11.76h). Our results indicate altered piroxicam pharmacokinetics in the conditions of acute liver injury
