Abstrakt | Phospholipase A2 (PLA,) hydrolyzes the sn-2 fatty acyl group of glycerophospholipids to release a free fatty acid and a lysophospholipid. Over the past decade, it has become clear that mammalian cells not only express a variety of intracellular enzymes, but also a diverse set of secreted phospholipases Az (sPLA2s). While PLA2s are generally considered as key enzymes which control lipid mediator production, the function of the 10 distinet sPLA,s cloned so far remains ill-defined. sPLA,s are also found in abundance in snake venoms and display a wide array of toxic effects including neurotoxicity and myotoxicity. Through the study of venom sPLAZS, two types of specific membrane receptors (N and M) have been identitied in various tissues. Of physiological relevance, the M-type receptor can bind with high affinities to several mammalian sPLA2s, making it likely that mammalian sPLA2S are endogenous ligands of the receptors initially identified with venom sPLA2S, and that the physiological function of the mammalian sPLA2s is not limited to their catalytic activily. | |
Izvleček | Fosfolipaze A2 (PLA2) hidrolizirajo sn-2 acilno skupino maščobne kisline glicerofosfolipidov. S hidrolizo se odcepita prosta maščobna kislina in lizofosfolipid. V zadnjem desetletju smo spoznali, da sesalske celice poleg ekspresije različnih intracelularnih encimov, ekspresirajo tudi različne fosfolipaze A2. Medtem ko PLA2 smatrajo kot ključne encime pri metabolizmu lipidov, pa ostaja vloga osmih kloniranih PLA2 slabo poznana. PLA2 so tudi pomembna komponenta kačjih strupov, nekatere od njih kažejo različne toksične učinke, kol sta npr. nevrotoksičnost in miotoksičnost. Z raziskavami toksičnih kačjih PLA2 smo uspeli v različnih tkivih določiti dva tipa rcceptorjev, tip N in tip M. Slednji lahko veže z veliko afiniteto več sesalskih PLA2, M receptor je kandidat za vezavno mesto endogenih sesalskih fosfolipaz, zaradi česar sklepamo, da te fosfolipaze nimajo zgolj katalitičnih lastnosti. | |
