Abstrakt     Heart Protection Study was randomized clinical trial with the aim to assess reliably the effects on mortality and major morbidity of cholesterol-lowering therapy and of antioxidant vitamin supplementation in a wide range of different categories of patients at high risk for vascular events. The patients with coronary disease, occlusive disease of non-coronary arteries, diabetes mellitus or treated hypertension and high-risk patients with baseline blood total cholesterol of 3,5 mmol/1 or greater of both gender aged 40 to 80 were eligible for the study. Between July 1994 and May 1997, 15 454 men and 5082 women were randomized in a group receiving simvastatin 40 mg daily or matching placebo tablets and, in a "2x2 factorial" design, antioxidant vitamin (600 mg vitamin E, 250 mg vitamin C and 20 mg beta-carotene daily) or matching placebo capsules. Patients were followed for at least 5 years. The results evaluating early safety and efficacy after mean 25 months of follow-up have already been published showing good tolerance of both treatment regimen and substantial effects on blood lipid and vitamin levels. The final results of the study which will be presented this year should provide reliable data about the effects of cholesterol-lowering and of antioxidant vitamin supplement on all-cause or cause-specific mortality and major morbidity in individuals for whom uncertainty remains about the balance of benefits and risks of these treatments.
