Abstrakt | A method is described for the determination of residues of dimetridazole in porcine and poultry muscle tissue. After the tissue has been homogenized in dichloromethane, the extract is evaporated and redissolved in hexane. Dimetridazole is extracted into buffer solution. Further purification is done with SPE on C-18 column. Dimetridazole is eluted with buffer and methanol mixture, concentrated and redissolved in mobile phase. Quantification is done with HPLC on reversed phase column (C-18) using acetonitrile, water, acetic acid and thriethanolamine as mobile phase with UV detection at 320 nm. The limit of detection is 5 pg/kg and the limit of quantification is 10 pg/kg with the coefficient of variation 11 %. Linear calibration graph was obtained in the range 0 - 50 micro g/kg with the correlation coefficient 0,9997. The situation regarding residues of dimetridazole in Slovenia from 1991 to 1997 is shown. | |
