Abstrakt     Cathepsins B and L are lysosomal cysteine proteases implicated in various stages of neoplastic progression. Elevated mRNA, protein and activity levels of cathepsin B and cathepsin L in tumour cells were found associated with increased invasion and metastasis formation in many types of cancers. In the present study the activity and concentrations of cathepsin B and cathepsin L were measured in cell lines derived from neuroepithelial (astrocytic) brain tumours of different histological type and grade of malignancies (astrocytoma, anaplastic astrocytoma and glioblastoma multiforme). Significantly higher concentration (up to 5.5 fold) of cathepsin L and progressively increased activity were observed in highly malignant glioblastoma multiforme compared to less malignant anaplastic astrocytoma and astrocytoma. Similar was found for cathepsin B where higher concentration (up to 9 fold) and progressively increased enzyme activity were found in more malignant astrocytic cells. Concentration of stefin B, endogenous inhibitor of cysteine proteases, was also slightly increased in anaplastic astrocytoma and glioblastoma multiforme compared to astrocytoma. Stefin A concentration was below the detection limit in all cell lines studied. Glioblastoma multiforme cells showed higher (2 fold) invasion potential compared to low-grade gliomas. Synthetic inhibitors of cysteine proteases partially inhibited invasion of anaplastic astrocytoma cells. The most potent was CA074, selective extracellular inhibitor of cathepsin B (83% of inhibition) followed by CA074Me selective intracellular inhibitor of cathepsin B (65% of inhibition). E64, general inhibitor of cysteine proteases inhibited invasion up to 57%. (Abstract truncated at 2000 characters)
