Abstrakt     Antigen presentation of bacterial proteins in infected tissues and influence of IFN-gamma on antigen presentation remains to a large degree unresolved. Balb/c mice (WT) and IFN-gamma gene-knock-out mice (GKO) were infected with Listeria monocytogenes. A new method, an ELISPOT-based antigen presentation assay, was used to detect four L. monocytogenes CD8 T cell apitopes. The antigen presentation proved to be a highly dynamic process. Five hours after the infection all four epitopes were present in similar amounts, but already 48 hours after the infection the amount of the presented epitopes differed significantly. 48 hours after the infection the amount of the epitopes correlated with the stability of the complexes that these epitopes form with MHC class I molecules. The combination of the ELISPOT-based antigen presentation assay and immunomagnetic separation of spleen cells enabled us to determine the antigen presentation on isolated specialised antigen presenting cells. 48 hours after the infection the antigen presentation pattern of the four epitopes on dendritic cells and macrophages differed between WT and GKO mice. Further analysis in vitro showed that IFN-gamma increases, decreases or does not effect the generation of different epitopes. The observed differences in the amounts of generated epitopes correlated well with the differences observed in the frequencies of epitope-specific T cells in WT and GKO mice.
