Abstrakt     Background. Ischaemia / hypoxia-reoxygenation induced injury of the vascular endothelium and smooth muscle cells alters the contractile response of the coronary artery. Preconditioning preserves cardiomyocytes against the ischaemia-reperfusion injury and the contractile function is restored after ischaemia-reperfusion period. Aim. The aim of our study was to determine the influence of 60-min hypoxia / 20-min reoxygenation and of preconditioning on the contractile response of the porcine coronary artery in vitro after hypoxia-reoxygenation. Hypothesis. We tried to confirm the hypothesis that contractile response of the porcine coronary artery can be preserved to a larger extent after hypoxia-reoxygenation with the use of hypoxic and pharmacological preconditioning. Methods. Rings (3-5 mm wide) of the left descending porcine coronary artery were placed in 10-m1 organ baths, filled with oxygenated Krebs-Henseleit (K-H) solution at 37 degrees C. Hypoxia was performed by the aeration of the K-H solution with a mixture of 95 % N2 and S % CO2. Contractions were measured with mechanical-electrical transducer. Contractile responses to 20 mM KCl before and after hypoxia-reoxygenation were compared. We compared the contraction after 60-min hypoxia to the contraction after 45-min hypoxia and to the contraction after 60-min hypoxia following the preconditioning. The hypoxic preconditioning consisted of two periods of 5-min hypoxia and 10-min reoxygenation. Pharmacological preconditioning was performed with bradykinin, adenosine, noradrenaline, adrenaline, papaverine, angiotensin II and acetylcholine. We also compared the hypoxic contraction and relaxation between arteries with endothelium and without it. Results were expressed as mean +- standard error of the mean.
