Abstrakt     Background: The quantity of DNA in the nuclei of the cells measured by cytometry is expressed in arbitrary units and graphically presented by histogram. The DNA value of the sample is compared with DNA value of normal cells and expressed by the DNA index. The DNA index indicates a relative level of deviation from normal diploid cells that have, by definition, a DNA index 1. The selection of appropriate reference cells is of crucial importance for a proper determination of DNA ploidy. In the fluid of the ovarian follicle that has been aspirated for the purposes of IVF, there is an abundant quantity of granulosa cells that are usually rejected after the removal of oocyte. The aim of our study was to find out whether these granulosa cells (GC) could be applied in the standardization of the measurements of DNA ploidy in fresh, not fixated cells by image- and flow-cytometer. We therefore analyzed the cellular content of follicular fluid , its proliferation characteristics and DNA content of the GC. Methods: In the first series we collected all aspirated follicles of one woman into the same sample (mixed follicles). We analyzed the follicular fluid sediments of 78 women with gonadotropin stimulated menstrual cycle, 10 women with natural menstrual cycle, and natural follicles obtained from the surgically removed ovaries of 8 women. In the second series we analyzed 88 samples of separated follicles from 10 women with gonadotropin stimulated menstrual cycle. The samples for the analysis of morphological and proliferation characteristics were centrifuged. From each sample, 2 air-dried smears and 4 cytospins were prepared which were fixated in Delauney. The air-dried smears were stained by Giemsa, whereas the Delauney-fixed cytospins were stained by Papanicolaou. One cytospin of each sample was also stained by MIB-1.
