Abstrakt     Adenocarcinoma of the stomach is a frequent malignant disease in Slovenia. Gastric cancer is a consecuence of a multi-step carcinogenic process and epigenetic changes, which include activation of oncogenes and inactivation of tumor suppressor genes. Different histological forms aswell as different etiologies point to two different genetic pathways for well differentiated and poorly differentiated gastric cancers. DNA mismatch repair is a defence mechanism, which can prevent the development of cancer in many people, and decreased efficiency of repair is an importent event in carcinogenesis. Our study included evaluation of microsatellite instability (MSI) on MMR associated locuses and loss of heterosigosity (LOH) on locuses associated with the following tumor suppressors: APC, nm23, Rb and p53. We succeded in multiplex-PCR and duplex-PCR for some microsatellite loci and uniplex-PCR for the rest. MSI and LOH was correlated with clinicopathologic factors of 74 patients, which we obtained from the Gastroenterologic clinic of the Clinical Center in Ljubljana. LOH was found in 53% of the informative samples, from which 22% were LOH-H and 31% LOH-L. We found a correlation of MSI with 5% LOH-L and 5% LOH-H tumors. LOH-H and MSI-H were not associated. For APC we found LOH in 36%, for p53 in 37%, for Rb in 24% and for nm23 in 32,5% of informative samples. MSI was found in 30% of the samples, from which 12% were MSI-H and 18% MSI-L. LOH-H status was associated with intestinal type cancer (70%), larger nodal invasion, diagnosis in higher TNM status and less differentiation. LOH-N status was associated with infiltrative growth and ulcering, infiltrating type of tumors. (Abstract truncated at 2000 characters).
